چکیده

It has long been recognized that the usual methods for cleavage of steroidal conjugates by strong acids may effect various transformations (1). In an earlier publication (1) one particular alteration, the conversion of a 3&hydroxy-17-ketosteroid to a non-digitonin-precipitable (“a”)-17-ketosteroid’ by standard HCl hydrolysis was studied. This conversion could be avoided by use of the hydrolytic procedure devised for sodium dehydroisoandrosterone sulfate in water or urine by Talbot et al. (2) with barium chloride in acetate buffer, pH 5.8. The present paper reports a series of experiments designed to determine (a) the essential factors involved in this hydrolytic procedure and (b) the nature of the conjugated 17-ketosteroids of normal male urine which are hydrolyzed by this treatment.

A Method for the Rapid Hydrolysis of Urinary Glucuronoside Metabolites of Testosterone.

..A.LMOST ALL the neutral urinary 17-ketosteroids are conjugated with either sulfuric or glucuronic acids. These conjugates can be cleaved by strong acids at elevated temperatures (1), or by solvolysis in either ethyl acetate or tetrahydrohiran, each containing perchioric acid (2). Selective hydrolysis of steroid-glucuronosicles is achieved with $-glucuromi_ idase, although in some 1i1#{149}ill...

Evaluation of hydrolytic products of 17-ketosteroids by means of gas-liquid chromatography.

In the course of evaluating urinary 17-ketosteroid (17-KS) gas-liquid chromatographic patterns, a number of peaks with retention times shorter than that of 17-KS were constantly observed. This backgroundmaterial was found to be degradation products of 17-KS produced by HCI hydrolysis. A study with pure 17-KS as a mixture and as individual steroids was used to identify the steroids responsible f...

The effect of formalin and the duration of acid hydrolysis on free and conjugated urinary 17-ketosteroids.

The recovery of five free ketosteroids, three glucuronides, and six sulfates, added to water or urine and subjected to acid hydrolysis for various lengths of time, was investigated. Dehydroepiandrosterone (DHEA) in all its forms was most susceptible to destruction. The behavior of glucuronides and sulfates differed, and no single set of conditions provided equal recovery for all conjugates. A c...

Quantitative paper chromatography of conjugated 17-ketosteroids in plasma.

The determination of urinary 17-ketosteroids has limited applications in the understanding and diagnosis of the nature of endocrine disorders associated with mild hirsutism and virilism in the female, because a very large number of these patients excretes 17-ketosteroids well within the normal range. Recent experience of Finkelstein, Forchielli and Dorfman (1) suggests that in these cases the b...

DESTRUCTION OF 3-&HYDROXY 17-KETOSTEROIDS DURING HYDROLYSIS WITH HYDROCHLORIC ACID* BY JOEL BITMANt

Acid hydrolysis has long been used to free the urinary 17-ketosteroids from the water-soluble combinations in which they are excreted. Both hydrochloric acid (13,5,11) and sulfuric acid (15)’ have been employed, for this purpose. The acid treatment, while effecting relatively complete hydrolysis, may also result in destruction (13, 5, 6,3, IS), chlorine substitution (2), dehydration (20,4, 12),...
